Abstract
Enzymes are required in order to create bioengineered heparin. N-Sulfo heparosan (NS), a heparin-like structure, undergoes multiple enzymatic reactions to yield a heparin generic with similar chemical and biological activity. Two of the enzymes: C-5 epimerase and 2-O-sulfotransferase are used in the first step of the bioengineered heparin process. Unfortunately, this step gives low yields of the converted product (NS to NS2S) requiring a 1:1 substrate to enzyme concentration to see appreciable effects. Higher concentrations of enzyme are used to achieve yield that are deemed acceptable to move on subsequent enzymatic steps.
